Son bac en poche en 1996, Sophie Guillemin exerce plusieurs petits boulots, notamment figurante à Euro Disney et serveuse dans la restauration rapide. En 1998, remarquée par Cédric Kahn, elle décroche le rôle principal de L'Ennui qui lui vaudra une nomination aux Césars dans la catégorie meilleur espoir féminin. Elle est remarquée à l'époque pour ses formes généreuses, et considérée comme une « révélation ».
